ANA (Antinuclear Antibody) Test: Unmasking Lupus & Autoimmune Diseases

Autoimmune diseases are among the most frustrating medical mysteries on earth. One month you have aching fingers; the next month you develop a strange red rash across your cheeks in the sun; then comes extreme exhaustion that no amount of sleep fixes. Because symptoms flare up and disappear unpredictably, patients often spend years visiting dermatologist after neurologist before getting answers.

When the immune system loses its ability to recognize your own cells and begins attacking the nucleus of your healthy tissues, it produces Antinuclear Antibodies (ANA). An ANA Test by Immunofluorescence Assay (IFA) is the gold-standard starting point for investigating autoimmune disorders like Lupus (SLE), Sjogren's syndrome, and Scleroderma.

Why IFA is the Gold Standard Method

There are cheap rapid enzyme tests (ELISA) for ANA, but rheumatologists overwhelmingly recommend the Indirect Immunofluorescence Assay (IFA) on HEp-2 cells. This method not only tells you if antibodies exist, but it also reveals two critical pieces of data:

  1. The Titer (Dilution Strength): Expressed as a ratio like 1:80, 1:160, 1:320, or 1:1280. The higher the second number, the stronger the concentration of autoantibodies.
  2. The Pattern (Nuclear Architecture): How the antibodies light up under ultraviolet microscopy:
    • Homogeneous / Diffuse: Commonly linked with Systemic Lupus Erythematosus (SLE).
    • Speckled: Associated with Sjogren's, Mixed Connective Tissue Disease (MCTD), and Scleroderma.
    • Centromere: Typical for limited cutaneous systemic sclerosis.
ANA TiterClinical Interpretation
Negative (< 1:80)Autoimmune disease unlikely; look for other causes.
1:80 to 1:160Borderline / Weak Positive (Can occur in 5% of healthy individuals).
1:320 and AboveClinically Significant Positive; requires specialized Autoimmune Profile (dsDNA, ENA panel).

Stop guessing. If you suspect an autoimmune condition, BookMyPatho offers advanced ANA IFA testing and comprehensive autoimmune panels with expert home collection in Delhi NCR.
